My advice
Book online before you go. It skips the ticket-desk queue, though the real crush is inside the small rooms, not at the till. The DDR Museum is privately run, so a Museum Island ticket or the Museum Pass Berlin does not cover it.
Where this shape comes from. Nobody publishes hour-by-hour visitor counts for Berlin museums, so this is not measured data and I will not pretend it is. It is my own read of the day from walking this part of Mitte several times a week: when the coaches unload, when the school groups arrive, when the rain pushes everyone indoors. Treat it as a guide's estimate, not a live queue counter.
